Panel A. pSMART® ChimeraFree vectors (clone up to 8-10 kb). The pSMART vectors are available in either high copy or low copy versions. G-tailed insert DNA is ligated to the C-tailed vector termini, ensuring only one insert per vector. Ori, origin of replication; Kan, Kanamycin resistance gene. Approximate positions of sequencing primers and CloneSmart transcription terminators (T) are indicated. The low copy vector includes ROP, Repressor of primer (lowers plasmid copy number).

Panel B. pJAZZ ChimeraFree linear vectors (clone up to 30 kb). The copy number is ~4-10/cell prior to induction; it is increased by approximately 10-20 fold by a simple chemical induction step in the included BigEasy®-TSA Cells. (1) Before processing, the pJAZZ vector (12.9 kb) contains a lacZ stuffer fragment. (2) After processing, the vector has 3’-C tails and dephosphorylated ends. G-tailed insert DNA is ligated to the C-tailed vector arms, ensuring only one insert per vector. The left arm is 10 kb and the right arm is 2.2 kb.
